USD"],"formats":[],"version":null,"rightsList":[],"descriptions":[{"description":"With this Pilot CPPRA we propose to develop a new program to reduce and prevent commercial tobacco use among African Americans returning to San Francisco Bay Area communities from incarceration. People in and returned from prison are four times as likely to smoke as nonincarcerated people. Black men who were ever imprisoned are nearly twice as likely to smoke as Black men who were never imprisoned. Studies show that most people returning from prison want to quit, but face many challenges in re-entering society, and these challenges can make it harder to quit smoking. People may not know about or have confidence in existing cessation programs, and these programs may not be suited to this community. Our pilot project brings expertise in tobacco research and intervention from Dr. Juliet Lee and the Pacific Institute for Research and Evaluation (PIRE California), together with community expertise from Mr. Palmer and Life After Next, a community-based organization in the San Francisco Bay Area working with African Americans and Men of Color returning from incarceration. Life After Next provides advocacy, mutual support, and connections to services, in particular support for housing and mental wellness and is well-connected to the Black return community and the network of providers that serve the community. Despite this women of reproductive age and pregnant women are still affected by harmful chemicals in cigarette smoke. Tobacco smoke may be inhaled directly by smoking (firsthand) or indirectly by being around others who smoke (secondhand) or being in areas where someone has smoked (thirdhand). Tobacco smoke contains cancer-causing chemicals called polycyclic aromatic hydrocarbons (PAH). PAH exposure is linked with low birth weight in babies, early birth of babies, and difficulty getting pregnant in women. Benzo(a)pyrene (BaP) is a specific PAH that is found cigarette smoke. BaP harms the ovaries, which contain the limited number of oocytes. Oocytes eventually grow into eggs that a woman needs for a healthy pregnancy. In order for an oocyte to become an egg it must mature through meiosis, a special type of cell division specific to eggs and sperm. Problems with meiosis lead to unhealthy oocytes that make it difficult for a woman to become pregnant and also contribute to birth defects. Mouse research from the Luderer laboratory shows that BaP exposure during pregnancy greatly reduces the number of oocytes and disturbs the expression of meiosis genes in developing ovaries. However, the specific parts of meiosis that are affected and whether the quality of the oocytes that survive and eventually become eggs is affected are unknown. To address these gaps in knowledge I will be collecting oocytes from developing and adult ovaries of mice exposed to BaP during pregnancy. I will check these oocytes to find out how BaP damages an early meiotic process called synapsis which is needed for the exchange of DNA between chromosomes and cohesion which is a chromosome or DNA stabilizing structure. This research is not only important because a healthy ovary is important for pregnancy. Recognizing the urgency of curbing the tobacco epidemic and promoting health equity, California localities such as Oakland (2018), San Francisco (2019), and Los Angeles (2020) implemented comprehensive bans on menthol and flavored tobacco sales, culminating in a state-level ban in 2023 through Senate Bill 793 and Assembly Bill 935; 138 CA local jurisdictions in 28 counties had adopted local bans that somewhat regulated the sales of flavored tobacco products. Between 2015-2023, the local flavor bans covered 46.5% of the state population. However, the impact of these flavor bans remains mixed. A 2023 study of discarded tobacco product packaging in 10 California cities found that menthol and menthol workaround products accounted for 21.1% of the total packs collected, suggesting persistent use despite the bans. Evidence also indicates the emergence of illicit markets for menthol-flavored products and possibly inadvertently shift towards traditional smoking in some populations. Given these developments, there is a need to evaluate California’s flavor bans, leveraging newly available data and focusing on health equity. My proposed studies will assess the impact of state and local comprehensive flavor bans on tobacco use prevalence among children, teens, and young adults (\u0026lt; 26 years), using national (Population Assessment of Tobacco and Health Study) and California surveys (California Healthy Kids Survey). These studies will 1) Reevaluate local flavor bans' effects on tobacco use (e.g., cigarettes and e-cigarettes) incorporating extended time frames, newly available data, and advanced quasi-experimental statistical methods. The newest and most popular devices contain a nicotine formulation called nicotine salts. Nicotine salts, created by adding an acid to nicotine, are considered more appealing than the original formulations with freebase nicotine. Long-term exposure in mice suggests freebase nicotine ECs promote the development of risk factors for cardiovascular disease, including blood pressure, but this has not been studied for nicotine salt. Due to their popularity, it is essential to determine EC health impacts and the influence of salt compared to freebase. Recent studies suggest that nicotine formulation influences EC emissions and their respective health impacts. Studies characterizing EC emissions have shown nicotine formulations have higher nicotine concentrations, but the implications for nicotine’s delivery into the body and subsequent effects are unclear. Studies comparing nicotine formulations' impact on nicotine metabolism and cardiac effects are limited and inconsistent. Therefore, it remains uncertain what role nicotine formulation has on the composition and characteristics of EC emissions and its impact on nicotine metabolism and the cardiovascular system. This project aims to answer this: 1) by characterizing the physical and chemical composition of EC emission with nicotine salt and freebase nicotine; 2) by measuring differences in metabolism and biological activity by exposing mice to either nicotine formulation; and 3) by identifying the nicotine formulation's influence on heart rate and blood pressure, risk factors of cardiovascular disease. We expect EC emissions from nicotine salts to contain higher nicotine levels than freebase nicotine and different compositions that influenc enicotine delivery. Nicotine salt exposed mice will have higher nicotine metabolism rates than freebase nicotine, in addition to increased signs of oxidative stress, higher heart rates, and blood pressure. Using more than one drug makes these harms even worse. It’s important to consider how youth use both tobacco and cannabis. Studies show that using cannabis can lead to more cigarette smoking and can make it harder to quit tobacco. LGBTQ+ teens face extra risks. They often start using tobacco and cannabis at younger ages, use these drugs more often, and have more problems from tobacco than their straight, not transgender peers. But, most research looking at using tobacco and cannabis has not focused on LGBTQ+ teens. Research has mainly looked at LGBTQ+ adults instead, but this ignores the different ways that teens may use drugs. Also, surveys that researchers use today don’t always tell us if teens are using both drugs at the same moment or just over the same time period. These two patterns can happen for different reasons so it’s important for researchers to tell them apart. Once researchers learn more about how LGBTQ+ teens use tobacco and cannabis, we can create better programs to help them stop or use less. In this project, we will first look at two sources of data. The first source is surveys completed by LGBTQ+ teens in California five times each day for two weeks. The second source is from a large, national survey called PATH that looks at tobacco use in the U.S. We will use what we learn from these surveys to do interviews with LGBTQ+ teens in San Diego. In these interviews, we can learn more about what leads them to use both tobacco and cannabis. This research will help show how often LGBTQ+ teens use both tobacco and cannabis. It will also point out the reasons why LGBTQ+ teens use them. Latinx youth make up almost half of all young people in the state and are more likely to use tobacco than some other groups. Yet, they often struggle to get support to quit and are left out of research. Many also don’t have access to programs that match their language and culture. This project focuses especially on Latinx youth who prefer to speak Spanish. Spanish-speaking Latinx youth who came to the U.S. without parents or guardians face even more challenges. They may not speak English well, may not have health insurance, and may be dealing with stress and trauma from moving to a new country. These issues can make it easier for them to start or keep using tobacco. If they are also homeless or don’t have a stable place to live, the risk is even higher. In fact, up to 70% of youth without housing use tobacco. This study will create a program to help Spanish-speaking Latinx youth without stable housing. The program will be built with help from youth and community partners. One goal is to learn how to design the program with the people it’s meant to help, not just for them. Another goal is to compare how well the program works for Spanish-speaking youth and English-speaking youth. By listening to youth and community members, the project will learn what parts of the program work well and what needs to change. The goal is to build a better program that meets the real needs of these youth. Through this project, Ms. Lopez will learn how to create health programs and work closely with the community. She will also do research that helps people who often don’t get the care they need. A significant factor that contributes to NSCLC is the exposure to cigarette smoking, which have been shown to induce tumor metastasis, the process facilitating the spread of the cancer cells. This tumor metastasis is initiated by a biological phenomenon called epithelial-to-mesenchymal transition (EMT) of cancer cells. Tobacco smoking has been shown to trigger EMT and lung cancer metastasis. Metastatic lung cancer has an extremely low survival rate, and there is a need to explore new therapeutic targets. Cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling pathway can sense DNA damage caused by smoke products and is suggested to be critical for smoking induced NSCLC. Moreover, recent studies have suggested the role of cGAS-STING pathway in promoting EMT in cells. To understand the underlying mechanism, we will investigate how acute and chronic exposure to tobacco smoke products and nicotine impact the dynamics of cyclic GMP-AMP (cGAMP), the key signaling molecule synthesized by cGAS. We will further evaluate the role of cGAMP in regulating EMT of NSCLC cells exposed to tobacco smoke products. SCLC is strongly associated with tobacco use, as ~95% of SCLC patients have a history of tobacco use. Chemo- and radiation therapy are standard of care for first-line treatment of SCLC, but patients quickly develop resistance, resulting in disappointing clinical outcomes. As the overall five-year survival rate for patients with SCLC is ~7%, new therapeutic approaches are urgently needed. Recent research demonstrated that SCLC cells express high levels of a family of enzymes involved in DNA repair called poly-(ADP)-ribose polymerases (PARP). Pharmacological PARP inhibition (PARPi) was successful in slowing growth in cell culture and mouse models of SCLC. However, early clinical trials with PARPi revealed insufficient responses. Therefore, identification of treatment combinations that improve the efficacy of PARPi in SCLC is important. Recently, several reports showed that deleting a specific helicase (a type of protein that unwinds DNA) makes cells more responsive to PARPi, identifying the helicase as a potential therapeutic target for the treatment in SCLC. However, there are no known drugs that target this helicase, and helicases are in general difficult to target. To overcome this, we utilised emerging protein degradation techniques, where we use small chemical molecules to hijack the cell’s own protein removal system to specifically target this helicase for destruction. Using this approach, we developed a novel small molecule helicase degrader, and our initial studies show that helicase degradation synergizes with the FDA-approved PARP inhibitor Olaparib in SCLC cells to decrease cell growth. Here, I will define how loss of this helicase sensitizes SCLC to PARP inhibition. Additionally, I will determine whether our compound has suitable properties to induce degradation in mouse models of SCLC. Lastly, as many SCLC patients continue to smoke even after diagnosis, and as nicotine exposure can reduce the effectiveness of chemotherapies, I will determine whether nicotine impacts the sensitivity of SCLC cells to PARPi and helicase degradation. This includes those who are gay, lesbian, bisexual, transgender, or nonbinary. Some SGM subgroups (like bisexual and transgender individuals) use e-cigarettes even more often. There have been efforts to lower use among these youth, but the differences still exist. This highlights a need to find out why. One possible reason is exposure to adverse childhood experiences (ACE). ACE are negative events that happen to people before age 18, like abuse or family problems. Research shows that young adults who have had more ACE are more likely to use e-cigarettes. SGM young people also report more ACE, like neglect. This might partially help explain the higher rates of use. The current way of measuring ACE may not fully show the unique problems that SGM youth face growing up. To help with this, researchers made a new tool called the SGM-ACE scale. This scale has 8 questions about stressful events related to being part of an SGM group. This includes bullying, family rejection, and religious trauma, before the age of 18. Higher SGM-ACE scores are linked to worse mental health (depression, anxiety). However, no one has used this tool to study e-cigarette use in the SGM population yet, or among SGM subgroups. This TRDRP study will thus explore how both ACE and SGM-ACE are related to e-cigarette use in SGM youth. It will also explore if these patterns are different for specific SGM subgroups. These findings can help us find better ways to support SGM populations. Despite recent advances, breast cancer remains the most common cancer diagnosis for women globally and within the United States. Triple negative breast cancers (TNBC), primarily found in women under the age of 40, account for 10-20% of all breast cancer diagnoses in the United States. TNBCs are noted for their aggressive reoccurrences and complex molecular heterogeneity. Traditional pharmacological treatments for breast cancer patients involve small molecules targeted against hormone receptors. However, the lack of hormone receptors in TNBC limits therapeutic options and therefore makes TNBC a critical field for pharmacological development. Previous work has demonstrated the role of alpha-arrestin domain containing protein 3 (ARRDC3) as a tumor suppressor in regulation of G-protein coupled receptor oncogenic signaling and tumor spread in TNBC cells. ARRDC3 is important for receptor degradation, but is decreased across various invasive breast cancer cell lines. In addition to GPCRs, oncogenic and inflammatory signaling is linked to TNF-alpha (TNF) activity on tumor necrosis factor receptor superfamily member 1A (TNFR1). TNF is a pro-inflammatory cytokine that signals through TNFR1 and increases migration and invasion of breast cancer cells. TNF is found at higher levels in the blood serum of smokers in comparison to serum TNF levels of non-smokers, and increases in a cigarette dose-dependent manner. Our work now investigates a role for ARRDC3 in nicotine-driven inflammatory signaling in invasive breast cancer. Our preliminary work reveals that ARRDC3 affects total TNFR1 expression on both untreated and TNF-treated invasive breast cancer cells. We have also found that ARRDC3 affects TNF-mediated signaling pathways and cytokine release from breast cancer cells. Yet, older adults, especially those aged 50 and up, also need help to quit smoking. There are a few studies on older smokers. In California, smoking rates are low in the general population. But 25% to 35% of Vietnamese American men still smoke. Older Vietnamese Americans may find it hard to quit. Language barriers can create challenges for them. These challenges make it hard for them to access quit-smoking services. Mental health issues, like stress and depression, often go hand in hand with smoking. Older Vietnamese Americans often find it hard to seek mental health help. Stigma and a lack of appropriate services make it even tougher. When they use smoking to cope with stress and anxiety, quitting becomes even more difficult. We need better policies and programs to help people quit smoking and support their mental health. This is especially true for older Vietnamese Americans. To tackle these gaps, I propose a study with two main parts. I will first talk to older Vietnamese Americans who smoke. I want to learn about their experiences and what support they need. I’ll also ask for their views on smoking and mental health. Next, I will talk to providers of quit-smoking support and mental health services. I want to see how these services can work together better. This research hopes to improve programs that assist older Vietnamese Americans in quitting smoking by also providing mental health support. What we learn may also help other groups facing similar challenges. This study and the training will help me become an independent researcher. PRISm patients generally present symptoms such as persistent cough, shortness of breath, and exercise intolerance. Moreover, they experience a higher incidence of comorbidities and a heightened risk of serious health complications, and about 40% progress to chronic obstructive pulmonary disease (COPD) within 5 years. For this reason PRISm is sometimes termed ‘pre-COPD’. COPD is the 4th leading cause of death in the US. PRISm is an understudied group and currently there are no effective treatments to improve their outlook. Recent studies of inhaler medications, effective in COPD to expand airways and reduce symptoms, showed that symptoms persisted in PRISm. Exercise training is the standard of care in COPD patients to alleviate symptoms improve quality of life and survival. However, the potential for exercise training to improve symptoms and exercise tolerance in PRISm have not been studied. The main objectives of this study are to: 1) determine the mechanisms and mediators of exercise tolerance in PRISm, including pulmonary abnormalities, leg muscle strength and body composition and 2) investigate the efficacy of a 12-week exercise training program to reduce symptoms and enhance exercise tolerance among individuals with PRISm. PRISm patients are a ‘forgotten group’ with very limited therapeutic options available. This research aims to bridge this gap by shedding light on the underlying mechanisms responsible for the symptoms and physical limitations associated with PRISm. Most lung cancers occur in patients who are current or prior smokers. In recent years, immunotherapy has proven to be an effective treatment for many lung cancer patients, especially for a subset of current/former smokers. However, some lung cancer patients fail to respond to this type of treatment. In particularly, tumors with mutations in the KEAP1 gene have been shown to respond poorly. KEAP1 mutations are highly enriched in lung cancers that develop in current/former smokers, and therefore overcoming the resistance they cause is a major unment need for this subgroup of lung cancer patients. The reasons behind this poor response to immunotherapy in this specific lung cancer subtype are not yet fully understood. We propose that the lack of response is due to differences in the type and characteristics of infiltrating immune cell between tumors with and without KEAP1 mutations. To test this idea, we will use cutting-edge techniques known as spatial transcriptomics and single cell RNA sequencing. These advanced techniques will allow us to examine which genes are turned on and off by immune and cancer cells and reveal their precise location within the tumor. Our research will uncover the variations in the immune responses between KEAP1 mutant and wild-type tumors.
