proposta de trabalho para o estágio de pós-doutorado envolve colaboração com a Medicines for Malaria Venture (MMV, www.mmv.org), uma organização de pesquisa e desenvolvimento sem fins lucrativos que O objetivo do projeto é investigar a síntese de análogos de moléculas alvo, enviadas pela MMV, como ponto de partida para descobrir um composto que obedeça aos critérios para candidatos finais da MMV, visando a prevenção e o tratamento da Malária. A duração deste projeto é de 12 meses. Para superar a acessibilidade limitada a moléculas e a capacidade reduzida de triagem, a MMV tem como objetivo catalisar a pesquisa para a descoberta de novas e pequenas moléculas ativas por via oral, com potencial para futuro desenvolvimento. Essas publicações constituem um enorme recurso para o avanço das atividades de descoberta de medicamentos contra a malária e redefinem as regras tradicionais relativas à propriedade intelectual. A pesquisadora de pós-doutorado deverá realizar a síntese e avaliação biológica das séries químicas enviadas pela MMV, a partir de seus programas de screening. Em transplantados renais, leva ao desenvolvimento de disfunção crônica do enxerto, com redução da taxa de filtração glomerular (TFG) e da sobrevida do enxerto. A aldosterona parece estar implicada na gênese e progressão da fibrose renal, por meio da ativação e do recrutamento de células inflamatórias, da adesão destas ao endotélio, da ativação de células residentes teciduais, da liberação de citocinas pró-inflamatórias e do aumento da produção e deposição de elementos da matriz extracelular. Antagonistas da aldosterona têm sido apontados como renoprotetores, ao reduzir a fibrose renal e a lesão vascular, com potencial impacto positivo na sobrevida do enxerto renal. O presente estudo clínico unicêntrico, randomizado, prospectivo e com intervenção aberta tem como objetivo avaliar o impacto do tratamento com antagonistas da aldosterona sobre parâmetros de fibrose e inflamação renais, medidos por meio de marcadores séricos e urinários, bem como por meio de análise histopatológica do tecido renal. Os pacientes submetidos ao transplante de rim de doadores falecidos serão randomizados, de acordo com as características dos doadores (padrão, critério expandido e lesão renal aguda), em três subgrupos, com base na intervenção: controle, espironolactona e finerenona. Os pacientes serão comparados prospectivamente por 12 meses, a fim de avaliar os efeitos dessas medicações sobre a função e a sobrevida do enxerto renal, bem como sobre marcadores histológicos e urinários de fibrose renal. A partir dos resultados obtidos neste estudo esperamos identificar, através de marcadores de fibrose renal, inflamação e estresse oxidativo, um efeito renoprotetor dos antagonistas da aldosterona (espironolactona e finerenona) nos subgrupos de pacientes tratados, quando comparados ao receptor não-tratado, do mesmo grupo de O uso da fibra de coco como precursor alia baixo custo, ampla disponibilidade e valorização de resíduo agroindustrial, contribuindo para soluções alinhadas à economia circular e à sustentabilidade de processos. O principal desafio científico consiste em estabelecer uma metodologia de síntese simples, reprodutível e de menor custo, capaz de gerar materiais com área superficial, porosidade e grupos funcionais adequados para favorecer a rota de dois elétrons da reação de redução do oxigênio. Para isso, serão avaliados parâmetros de ativação química e pirólise, incluindo tipo e concentração de ativador, temperatura e tempo de calcinação, com apoio de planejamento experimental para otimização das condições de obtenção. Todos os materiais sintetizados serão inicialmente triados por técnicas eletroquímicas, com destaque para medidas em eletrodo de disco-anel rotatório, espectroscopia de impedância eletroquímica e voltametria, permitindo selecionar as amostras mais promissoras para caracterizações estruturais e morfológicas complementares. The increasing availability of multi-ancestry WGS studies and biobanks provides unprecedented opportunities to discover rare coding and noncoding genetic variants, molecular endophenotypes, and regulatory mechanisms that influence common diseases. However, many discoveries remain out of reach because privacy restrictions and complex longitudinal or multi-phenotype study designs limit cross-biobank WGS analyses; xQTL and omics findings often cannot be translated into causal disease pathways; and disease-associated noncoding regulatory variants are difficult to prioritize in a phenotype-aware manner. This project will address these challenges with four aims. First, we will develop scalable and efficient meta-analysis frameworks for large-scale WGS studies and biobanks incorporating single-cell-sequencing-based functional data with complex study designs. Second, we will develop accurate and powerful tools for xQTL mapping and high-dimensional causal mediation analysis using omics data. Third, we will develop a phenotype-aware multimodal framework for pathogenicity scoring and prioritization of regulatory variants in common diseases by integrating allele-specific sequence representations, DNA-shape features, functional annotations, and phenotypic information. Fourth, we will provide user-friendly, end-to-end, and open-access tools adhering to Findability, Accessibility, Interoperability, and Reusability for Research Software (FAIR4RS) principles. We will apply the proposed methods to integrative analyses of the NHGRI GSP CCDG, NHLBI TOPMed, UK Biobank, METSIM, FUSION, and GTEx resources, validate discovery results using All of Us data, and benchmark regulatory variant prioritization using ClinVar and high-throughput functional genomics datasets from IGVF and ENCODE, including HepG2 base editing/CRISPRi and MPRA datasets. Our study aims to develop and validate a standardized questionnaire to measure the health care utilization (HUQ) of community-living adults with chronic diseases in Hong Kong. Literature review and expert interviews will first be conducted to develop a protocol of HUQ. For validation, the HUQ will be tested on older adults who have at least one chronic disease. The administration time and cost, test-retest reliability and validity of the HUQ will be analyzed. All data collected in this study will conform to Guidelines on Research Data Management and open to the community. El uso inadecuado de antibióticos ha contribuido al aumento de esta problemática. De este modo, ha surgido la necesidad de desarrollar y potenciar nuevos antibióticos con mayor estabilidad y espectro de actividad, como es el caso de los carbapenemicos. El imipenem es un carbapenemico ampliamente usado en la clínica como antibiótico de ultimo recurso para tratar infecciones por bacterias MDR. Sin embrago, existen mecanismos de resistencia al imipenem, entre los que se destacan la secreción de enzimas degradadoras del anillo β-lactamico, como las metaloenzimas de clase B, las cuales, son dependientes de Zn+2, agravando así el problema de resistencia. Estas enzimas son categorizadas en tres grupos: NDM, IMP y VIM, destacándose esta ultima por ser prevalente, exhibiendo multiples variantes y se encuentra ampliamente distribuida en el mundo. Para hacerle frente a este tipo de bacterias, los péptidos antimicrobianos (PAMs) han surgido como una opción debido a su alta capacidad bactericida, amplio espectro de actividad y baja respuesta de resistencia, como resultado de su mecanismo de acción inespecífico sobre la superficie de las membranas bacterianas. No obstante, mecanismos poco explorados como la quelación de metales, pueden llegar a tener una aplicación terapéutica promisoria, como por ejemplo, el secuestro de los iones de Zn+2 de las metaloenzimas para inhabilitar su función catalítica.  Por otro lado, la nanotecnología también ha llamado la atención para ser aplicada en el control de enfermedades, incluyendo a las infecciosas por bacterias MDR. El uso de nanosistemas para potenciar la actividad de un antibiótico convencionalmente usado en la clínica, es una alternativa atractiva. Las nanopartículas compuestas por polímeros biológicamente activos y no tóxicos como el quitosano, están siendo cada día más usados debido a la biocompatibilidad, baja toxicidad y a la eficiente liberación del antibiótico sobre la superficie bacteriana, mientras es protegido de las hostilidades del entorno, incluyendo la degradación enzimática. A esto se le suman, las propiedades antimicrobianas del quitosano. Para mejorar la funcionalidad de la nanoparticula polimérica, péptidos de interés funcional pueden ser adheridos a la superficie de la nanopartícula. Dicho lo anterior, en este trabajo se encapsulará imipenem dentro de nanopartículas de quitosano funcionalizadas en la superficie con distintos péptidos quelantes de Zn+2. La caracterización fisicoquímica de las nanoparticulas se estudiará por dispersión de luz dinámica (DLS) y potencial-Z. El perfil de liberación del imipenem se evaluará a pH de 7.4 usando el método de la bolsa de diálisis y los datos se ajustarán a modelos cinéticos de orden cero, uno, Higuchi, Korsmeyer-Peppas, Peppas-Sahlin, Baker y Hixson-Crowell. Asimismo, el perfil de liberación será caracterizado por el área bajo la curva y el tiempo promedio de residencia del imipenem. In response, a multi-year research effort developed the prototype, open-source Travel Risk In Pandemics (TRIP-X) simulation model to extend Safety Risk Management principles to respiratory and biological hazards in air travel. TRIP-X is intended to integrate ventilation, human behavior, operational, and microbiological factors using airport and aircraft data to quantify transmission risk, evaluate layered controls, and support evidence-based decision-making. Beyond pandemic preparedness, TRIP-X is planned to enable tailored protective strategies for repatriation and humanitarian operations—such as safely evacuating personnel during severe outbreaks without imposing unnecessary restrictions—while also optimizing outbreak and epidemic management to balance public-health protection with the continuity of air travel, commerce, and societal mobility. Additionally, it may support homeland security and threat preparedness by providing a framework for structured threat and vulnerability analyses of intentional biological releases, strengthening aviation safety, public-health coordination, and national defense readiness under a unified risk-management approach. The barrier to a HIV cure is rare, rebound competent proviruses in CD4 T cells that not cleared by antiretroviral therapy (ART) and can be maintained for decades. Targeting these latently infected cells is incredibility challenging. In addition to their rarity,  fully intact proviruses are outnumbered 100-fold by defective viruses. Moreover, HIV diversity between PWH further limits molecular approaches to detect provirus. Altogether, current methods to detect and characterize intact viruses are inefficient and involve costly screening of non-infected cells that vastly outnumber infected cells, limiting our understanding of the regulators of persistent infection and ability to confidently identify rebound competent virus in PWH on ART. In Aim 1, we propose a multi-step approach to recover, sequence and characterize rebound-competent virus in CD4 T cells in PWH. The key advance on other approaches is that only HIV infected cells are analyzed.  Aims 2 and 3 will examine overlooked but important biological questions about the HIV reservoir, each aimed at better understanding proviruses that are rebound competent. Despite advances in diabetes technology, most children and adolescents with T1D do not meet glycemic control recommendations, suggesting that many likely struggle to manage T1D effectively. Depressive symptoms further complicate one’s ability to engage in daily T1D management, with numerous studies linking greater depressive symptoms to less frequent blood glucose monitoring, more frequent insulin administration problems, and higher HbA1c values. Yet, most pediatric diabetes clinics are understaffed with mental health providers, fewer than 25% of youth complete an outpatient mental health visit after referral, and approximately half of those who initiate mental health services attend only one visit. Single‑session interventions (SSIs), however, offer a promising, scalable approach to delivering mental health support in low‑burden formats that fit the needs of youth and families managing chronic illness. The PI’s current K23 award (K23DK139431) focuses on developing an evidence-based depression SSI tailored for youth with T1D. The PI developed this SSI in partnership with diabetes providers and youth with T1D and is designed to be delivered by a trained clinician via telehealth. However, this model still requires scheduling, staff time, and clinical infrastructure. In contrast, a fully digital, self‑guided SSIs may dramatically expand access by eliminating reliance on clinician availability. Therefore, adapting the T1D-specific SSI into a fully digital, self-guided platform represents a logical and necessary next step to extend the reach of this evidence-based SSI without increasing demands on the behavioral health workforce. This R03 directly builds on the PI’s K23 by converting a clinician-delivered depression SSI, specifically tailored for youth with T1D, into a fully digital, self‑guided platform. The overarching aim is to establish the preliminary feasibility and acceptability of the digital SSI. The first aim is to digitize the T1D-tailored depression SSI developed during the PI’s current K23 project. The PI will translate the manualized, evidence‑based SSI into an engaging digital platform designed specifically for youth with T1D. This work will apply user‑centered design principles and include iterative prototyping with youth with T1D to ensure the interface is accessible, relevant to diabetes‑related stressors, and consistent with core therapeutic components of the clinician‑delivered intervention. The second aim is to evaluate the feasibility and acceptability of the digital SSI through a proof-of-concept trial with youth with T1D. The PI will conduct a single‑arm pilot study to assess the feasibility (e.g., recruitment rate, retention, and intervention completion) and acceptability (e.g., user satisfaction, perceived usefulness, qualitative feedback) of the digital SSI among a national sample of youth with T1D. Exploratory outcomes will include estimation of variability in depressive symptoms, diabetes distress, and glycemic outcomes to inform future trial design. Findings from this R03 will inform content refinement and optimization of a scalable digital SSI for youth with T1D prior to larger‑scale testing. Este projeto propõe o desenvolvimento de uma nova rota de fabricação para rebolos de diamante baseada na integração entre manufatura aditiva por Processamento Digital de Luz (Digital Light Processing - DLP), arquiteturas porosas projetadas por superfícies mínimas periódicas tridimensionais (TPMS) e manipulação acústica de partículas abrasivas por acustoforese. A hipótese central é que o posicionamento controlado de grãos abrasivos por campos acústicos, aliado ao controle geométrico da porosidade via TPMS, permitirá a produção de ferramentas com microestrutura determinística, resultando em melhor evacuação de cavacos, maior eficiência de refrigeração e desempenho superior na retificação. A metodologia envolve a formulação e otimização de suspensões resina-diamante por planejamento de experimentos (DoE) e caracterização da cinética de fotopolimerização por meio da Curva de Jacobs. Transdutores piezoelétricos serão integrados à plataforma DLP para induzir a organização espacial dos grãos abrasivos antes da cura de cada camada. A validação será realizada por ensaios de retificação de amostras de alumina (Al2O3) com monitoramento de forças de corte e emissão acústica. Espera-se demonstrar ganhos na relação G e na integridade superficial das peças. Meanwhile, half of all fertilized embryos are lost before clinical detection. These are not failures of modern medicine: they are the predictable outcomes of a fundamental biological conflict. Mother and offspring share only half their genes, so their genetic interests over resource allocation diverge – a tension encoded in the very hormones that sustain pregnancy. Within the TME, tumor, stromal and immune cell interactions drastically affect ovarian cancer behavior, enabling immune evasion and treatment resistance. Understanding the complex cellular interactions within the human ovarian TME is vital to enhancing ovarian cancer therapeutics. Indeed, the immune system is critical in the ovarian TME as the balance of immune-activating and immune-suppressing cells dramatically impacts patient outcomes. T cells within the ovarian TME are associated with improved patient survival while immunosuppressive cells, including T regulatory cells, are associated with decreased survival. However, immunotherapy (IO) trials in ovarian cancer have been disappointing with early phase checkpoint inhibition trials only yielding response rates of ~10%. Identifying immune-tumor-stromal interactions that modulate the balance of immune activation vs suppression is critical to improving IO in ovarian cancer patients. Tertiary lymphoid structures (TLS) are sophisticated, ectopic cellular neighborhoods that correlate with improved prognosis and superior IO response. Indeed, a combined signature for TLS and CD8+ T cells correlates with increased patient survival. Given these clinical data, organization of immune cells into TLS is critical for the function of immune cells within tissues. Specifically, immune responses require interactions between different immune cell types, timing and coordination between these cells, and pro-survival cues to maintain immune cells long-term. Tumors develop in ectopic tissues over the course of many years; thus, it is unclear how responses are maintained within tumors. This is a central question in IO, as it is widely accepted that efficacy relies on the location of functional immune cells in tumors at the time of therapy. TLS would provide many or all the benefits for driving local immune responses in tumors by enabling and dictating critical interactions in the TME, thereby allowing for effective tumor killing. Given the therapeutic promise of TLS, we aim to understand B cell and TLS modulation by the tumor-stromal axis in ovarian cancer patients and physiologically relevant mouse models of ovarian cancer. Aim 1 will spatially evaluate key immune-tumor-stromal interactions with both robust spatial imaging and transcriptomics, ultimately building off our current preliminary data on ovarian cancer TLS. Aim 2 will measure the impact of one key stromal modulator, mesenchymal stromal cells (MSCs) on B-T cell crosstalk in vitro and in vivo. Our overall hypothesis is that there are differing immune and stromal mediators that influence TLS formation in ovarian cancer patients, and that these mediators can be enhanced or repressed for improved TLS formation and activity in the TME. However, mechanistically very little is known about HIV latency/persistence in myeloid cells. Notably, the presence of HIV-harboring myeloid cells in the CNS is documented to be the key factor contributing to CNS inflammation and promoting HIV-associated neurocognitive disorder (HAND) in patients.  Microglial cells are the main HIV reservoir in the CNS, yet there is a gap in the knowledge regarding our understanding of the molecular mechanisms that maintain HIV reservoirs in those cells.  We have shown the vital role of DNA-PK in supporting HIV transcription and latency-reactivation in both lymphoid and myeloid cells. The objective of this grant is to define the role of DNA-PK during HIV latency in microglia, outline the mechanisms through which DNA-PK cooperates with cocaine, and eventually test the clinical significance of in-human tested DNA-PKi in limiting HIV load and CNS reservoir in a mouse model.  We hypothesize that cocaine-induced DNA-PK promotes HIV transcription, leading to neurological impairment due to enhanced HIV replication and its gene products; however, clinically evaluated DNA-PK inhibitors can reverse this process. To test our hypothesis, In Aim 1, we will evaluate the cooperative impact of cocaine on DNA-PK-mediated upregulation of HIV transcription, replication, and latency-reactivation in microglia. In Aim 2, using human PBMCs, we will assess the impact of two highly specific and clinically evaluated DNA-PKi, Nu7441 and M3814, in restricting HIV transcription. In Aim 3, using a well-characterized HIV-infected humanized mouse model, we will test the therapeutic potential of in-human tested DNA-PKi in restricting HIV replication, reservoir, and viral rebound upon ART interruption. Upon conclusion, we will reveal the crucial role of DNA-PK in regulating HIV transcription and latency in microglia; delineate the physiological impact of in-human tested DNA-PKi, particularly in the context of cocaine and ART; and finally, we will establish the translational potential of DNA-PKi in limiting HIV replication, latent reservoir, and viral rebound upon ART interruption. Our rationale is that since HIV latency is primarily regulated at the transcriptional level, defining the precise mechanism(s) that regulate HIV transcription in microglial cells will offer new therapeutic opportunities to target HIV reservoirs in the CNS. The study will also provide a well-defined therapeutic target in the form of DNA-PK, and this new knowledge will be valuable for both basic and translational research. Proposed research is innovative because data will be confirmed in iPSC-derived microglia, besides validation in a humanized mice model. This contribution is significant since we will test the clinical relevance of DNA-PK inhibitors in controlling HIV and define the mechanisms regulating HIV transcription and latency in microglia. O objetivo é  classificar sessões como legítimas ou impostoras sem uso de senha explícita, utilizando modelos de classificação  binária (Decision Tree, SVM, Random Forest).   Este plano foi criado como atividade acadêmica da disciplina ESW-008/2026-2 - Engenharia da Informação (PECE - Poli USP).
