Tobbaco calmodulin-like protein rgs-CaM displays two EF-hands motifs that contains two putative calcium binding loops each. However, ITC studies showed that rgs-CaM displays only three calcium binding sites. Here, in silico model of rgs-CaM with calcium ions loaded in their four putative calcium binding loops shows that the calcium ion modeled in loop C escaped from the loop along the MD simulations highlighting that loop C is unable to coordinate calcium. In fact, aminoacids sequence comparison shows that loop C does not conserved any residue involved in calcium coordination.
