During Schistosoma mansoni infection, eggs released by female worms are embolized mostly in the intestine and the liver, where they induce granuloma formation which is sometimes followed by fibrosis. The intensive use of praziquantel for the treatment of schistosomiasis has raised concerns about the possible emergence of drug-resistant schistosomes. The search for alternative drugs has become a priority. The aim of this study was to histologically assess the antifibrotic potential of the ethyl acetate fraction from Ozoroa pulcherrima (EAOp) roots methanolic extract on S. mansoni-infected mice. The fraction at 100, 200 and 400 mg/kg was administered orally and daily to infected mice for 28 days, starting from 36th day post-infection. All mice were sacrificed at 65th day post-infection. Parasitological burden and histology of the liver were conducted. Hematoxylin and eosin (H&E) staining was performed to evaluate the inflammatory infiltration and the granulomatous reaction. Liver sections were also stained by picrosirius method for collagen evaluation. Giemsa staining method was also performed to visualize parasite eggs and the larvae inside it. The treatment produced significant reduction of the worm burden (p<0.01 and p<0.001) and the number of eggs accumulated in the liver (p<0.01 and p<0.001) at all doses. Histological analysis showed that after treatment of S. mansoni-infected mice with EAOp or praziquantel, inflammatory cells infiltration, the number and consistence of liver’s granuloma were reduced. This was characterized by an important reduction of collagen deposition around the eggs. This study is a histologically proof of the antifibrotic activity of Ozoroa pulcherrima ethyl acetate fraction and also supports the traditional use of this plant for the treatment of intestinal helminthiasis. EAOp could then be consider as an alternative or complementary therapy to praziquantel.
