Given the therapeutic potential of xenon, we aimed to study its effects after localized delivery to the brain using microbubbles. We designed xenon containing microbubbles stabilized by dibehenoylphosphatidylcholine (DBPC) and polyethylene glycol (PEG) attached to saturated phospholipid (DPSE-PEG5000). Using a pig model of TBI, these microbubbles will be intravenously injected, and ultrasound will be used release xenon at the level of the carotid artery. Control group will receive perfluorobutane containing microbubbles. Diffusion tensor imaging (DTI) will be performed to study white matter injury severity. at day 1 and day 5. On day five, animals will be euthanized and brain tissues of xenon treated animals will be analyzed for perivascular inflammation and blood-brain barrier disruption. Endothelial cell culture experiment will also be used to study the effect of xenon on changes in zona occludens-1 (ZO-1) after glutamate
